Latest RCT: Joint Health Support for Healthy Adults — Ryusendo to Exclusively Distribute Bioiberica’s Collavant® n2 in Japan
A new joint-health option featuring 40 mg/day of native (undenatured) type II collagen
Ryusendo Co., Ltd. is expanding Collavant® n2 in the Japanese market as the exclusive distributor for Japan. Collavant® n2 is a native (undenatured) type II collagen ingredient developed and manufactured by Bioiberica S.A.U. of Spain.
Collavant® n2 is derived from chicken sternum cartilage and contains undenatured type II collagen that retains the native three-dimensional structure of type II collagen. Unlike conventional collagen peptides, Collavant® n2 is characterized by an immunological mechanism known as oral tolerance, through which it supports joint health. Preserving the native structure of type II collagen is considered important to this proposed mechanism of action.
The recommended daily intake is only 40 mg. Despite this low inclusion level, the scientific evidence base continues to grow, including human clinical studies.

2026: Latest RCT published in healthy adults
In 2026, a new clinical study of Collavant® n2 was published in the international peer-reviewed journal Nutrition Journal.
The study was a randomized, double-blind, placebo-controlled trial involving 74 healthy adults who had not been diagnosed with knee osteoarthritis (OA) but experienced knee discomfort during physical activity.
Participants consumed either:
• Collavant® n2: 40 mg/day
• Placebo
for 180 days. Outcomes included the Knee injury and Osteoarthritis Outcome Score (KOOS), time to recovery from knee discomfort after standardized exercise, and urinary CTX-II, a biomarker of type II collagen degradation in cartilage.
In the Collavant® n2 group, improvements from baseline in the KOOS Pain, Symptoms, and knee-related Quality of Life (QOL) subscales were observed earlier than in the placebo group.
Among participants with relatively mild exercise-related discomfort at baseline, statistically significant between-group differences were observed at Day 180 in the change from baseline in KOOS Pain and QOL. Between-group differences were also observed in recovery time after exercise. In participants with greater discomfort at baseline, CTX-II decreased in the Collavant® n2 group but increased in the placebo group, with a between-group difference in the change from baseline.
These findings suggest that Collavant® n2 may have potential not only for traditional joint-health products targeting middle-aged and older adults, but also for a broader market of healthy individuals who experience joint discomfort during physical activity.

What is undenatured type II collagen?
Although products are often grouped together under the term “collagen,” their biological properties and proposed mechanisms of action can differ substantially depending on molecular structure.
Hydrolyzed collagen, or collagen peptides, consists of collagen that has been broken down into smaller peptides, and research has focused on the biological activity of absorbed peptides and related metabolites. In contrast, the undenatured type II collagen in Collavant® n2 retains the characteristic triple-helical conformation of type II collagen.
When consumed in small amounts while this native conformation is preserved, type II collagen is proposed to induce an immunological process known as oral tolerance through gut-associated lymphoid tissue. This mechanism is thought to help modulate immune responses associated with inflammation and tissue catabolism at the joint level.
Accordingly, Collavant® n2 represents a different approach to joint health from simply supplying large amounts of collagen as a structural nutrient.

Low-dose formulation: only 40 mg/day
The recommended daily intake of Collavant® n2 is 40 mg.
The ingredient is standardized to contain at least 6% undenatured type II collagen, providing at least 2.4 mg/day of native type II collagen at the recommended 40 mg daily intake.
The low inclusion level provides flexibility for a wide range of product concepts, including:
• Small tablets and capsules
• Combination formulas with other joint-health ingredients
• Sports nutrition products
• Products for active older adults
• Gummies, powders, beverages, and other dosage forms

Bioiberica’s expanding research platform in joint and cartilage health
Bioiberica, the developer and manufacturer of Collavant® n2, is headquartered in Spain and has a long track record of research and development in joint health and mobility.
Research on Collavant® n2 extends beyond human clinical trials to include articular cartilage, synovium, subchondral bone, MRI biomarkers, and gene expression. For example, animal studies have reported differences in cartilage, synovial tissue, subchondral bone, and MRI biomarkers when native type II collagen was added to glycosaminoglycans. Transcriptomic research has also investigated changes in gene expression related to extracellular matrix remodeling and homeostasis in chondrocytes.

From traditional joint care to the “active mobility” market
Joint-health supplements have traditionally focused on middle-aged and older consumers and on people already experiencing joint-related concerns.
Today, however, product concepts are increasingly expanding toward people who:
• Enjoy sports and regular physical activity
• Run or walk as part of their daily routine
• Want to support joint health while still healthy and active
• Are conscious of maintaining mobility as they age
With a recent RCT conducted in healthy adults, Collavant® n2 can be positioned not only in the conventional joint-health market for older consumers, but also in sports nutrition, active aging, healthy aging, and other emerging “active mobility” concepts.
